Disclosed is a clonal strain of acceptably attenuated vaccinia virus. It is isolated from cultured cells in which the New York City Board of Health vaccinia virus strain, ACAM 1000 (ATCC Deposit No. PTA-332 I), or a progeny thereof (æthe strainsÆ) has been cultured. The vaccinia virus is described to be able to induce a protective or therapeutic immune response against variola virus (the causative agent of smallpox) in a human. A method of obtaining a colonal strain of attenuated vaccinia virus for use as a vaccine is also described. The method comprises: (i) propagating the strains, or a progeny thereof in a cell culture system, and (ii) isolating from the cell culture system a clonal strain of vaccinia virus that has substantially the same virulence, immunogenicity, growth characteristics in culture, or restriction endonucleases digestion pattern as the strains. The virulence is determined by a rabbit skin test, or by a sucking mouse neurovirulence test.
