SOLUTION: The method of this invention calculates the indicator of the speed of Brownian movement of a first molecule, based on a fluorescence intensity of a reaction mixed solution that includes the fluorescent-labeled first molecule; a second molecule provided with a predetermined modified base and a third molecule specifically combined with the predetermined modified base. In such a case that the speed of the Brownian movement of the first molecule is determined to be lower than its speed, when the molecule is isolated independently, it is determined that the second molecule is bonded to the first molecule.
