A purified and isolated human plasma platelet-activating factor acetylhydrolase (PAF-AH) polypeptide fragment which is lacking up to the first twelve N-terminal amino acids of the mature human PAF-AH amino acid sequence set out in SEQ ID NO:8, his variant or variant fragment. An isolated polynucleotide encoding a human PAF-AH which is a DNA, a DNA vector comprising said DNA, a host cell stably transformed or transfected with DNA, a method of producing a PAF-AH comprising growing a host cell and a pharmaceutical composition comprising the PAF-AH. PAF-AH and said pharmaceutical composition are useful for the treatment of pleurisy, asthma, rhinitis, necrotizing enterocolitis, acute respiratory distress syndrome, acute pancreatitis, reperfusion injury, septicemia, preterm labor or neurological disease associated with HIV infection.
