1313595 Replicase for informationally active RNA D JACHERTZ 17 Aug 1970 [16 Aug 1969] 39477/70 Heading C3H A process for the preparation of a replicase for informationally active RNA comprises adjusting a cell-free system which has been obtained from immunologically competent cells to a pH in the range of 4À5 to 5, separating the precipitate so formed, washing and re-dissolving the precipitate, removing any remaining insoluble matter, fractionating the solution and separating the fraction having a distribution coefficient Kd in the range of 0À38 to 0À40 which contains the replicase; and if desired freezing said fraction for storage. "Immunologically competent" cells are those capable upon stimulation with an antigen of forming informationally active RNA; they may be peripheral leucocytes, spleen cells, lymph node cells, cells from the ductus thoracicus or peritoneal macrophages. The cells may be suspended in a buffer, cooled, frozen, crushed and centrifuged to produce a clear supernatant which is adjusted to pH 4À5 to 5 to obtain the precipitate containing the replicase. |
