Improved process of purification of streptomycin wherein the whole fermentation broth containing streptomycin is adsorbed on a fixed bed column of weakly acidic carboxylic acid cation exchange resin (e.g. a cross linked copolymer of acrylic or methacrylic acid). The bed is then washed with about one bed volume of a recirculating or single pass solution of an amino acid type chelating agent (e.g. ethylenediamine tetraacetic acid sodium salt solution, about pH 8) for about 1-3 hrs. The resin is then treated with carbonic acid at a rate of about 1-5 bed volumes per hour under a pressure of 5-50 p.s.i. for 3-10 hrs. The purified streptomycin is then eluted from the resin with a moderately concentrated mineral acid (e.g. H2SO4) at a slow linear flow rate until the pH of the pooled effluent drops to about 5-5.5. The streptomycin (e.g. as sulphate) is then isolated from the eluate by e.g. decolourization and lyophilization. Single step process which thus prevents loss of material due to repeated transfers. |
