PROBLEM TO BE SOLVED: To provide a variant immunoglobulin, especially a humanized antibody polypeptide with a method for producing and use of it, further a consensus immunoglobulin sequence and a structural model. SOLUTION: The variant immunoglobulin is obtained by obtaining an import antibody variable domain for making a humanized antibody comprising amino acid sequence of a non-human, import (transferred) antibody and a human antibody, and an amino acid sequence of a variable consensus domain, substituting an imported CDR amino acid sequence in place of a corresponding human CDR amino acid sequence, and determining if the non-homologous import amino acid residue is reasonably expected to have at least one of the following effects: 1. non-covalently binds antigen directly; 2. interacts with a CDR; and 3. participates in the VL-VH interface; substituting that residue for the corresponding amino acid residue in the consensus antibody FR sequence.
