PROBLEM TO BE SOLVED: To provide a new technique having a function (transfer vesicle escaping ability) for overcoming the transfer vesicle escape which is the maximum barrier in a travel from the outside of a cell to the inside of the cell nucleus and ensuring efficient transfection when a desired gene is transferred into the cell.
SOLUTION: A nucleic acid structure is formed by binding a complex composed of a nucleic acid substance containing a gene to be transferred into the cell and a cationic polymer to a transfer vesicle escape-promoting protein. A method for transferring the gene into the cell comprising a step of bringing the nucleic acid structure into contact with the cell is provided. In a preferred embodiment, the nucleic acid substance containing the gene to be transferred into the cell is a plasmid DNA, and the plasmid DNA is complexed with a biotinylated cationic polymer such as a biotinylated polyethyleneimine. In addition, the plasmid DNA is bound to diphtheria toxin T-domain-fused streptavidin as the transfer vesicle escape-promoting protein.
COPYRIGHT: (C)2009,JPO&INPIT |
