Provided are methods of assessing the absence or presence of a tauopathy in a subject comprising characterizing the microglial activation state in a neurological tissue of the subject prior to the clinical manifestation of said tauopathy. Also provided are methods of assessing the absence of presence of a tauopathy in a subject comprising determining the absence or presence of synaptic pathology in a neurological tissue of the subject prior to the clinical manifestation of said tauopathy. Also disclosed are transgenic PS19 mice expressing human P301S mutant tau, and methods for identifying an agent that modulates the onset or progression of a neurodegenerative tauopathy.
