A method of manufacturing an 3-O-substituted ascorbic acid derivative is provide to reduce a manufacturing cost by removing a process of introducing a protection period and a process of removing the protection period and to establish easy mass production. A method of manufacturing an ascorbic acid derivative comprises steps of: manufacturing an ascorbic acid ammonium salt by reacting the ascorbic acid of a chemical formula 2 and an ammonium salt of a chemical formula 3; manufacturing an 3-O-substituted ascorbyl ether of a chemical formula 1 by reacting the ascorbic acid ammonium salt and a compound of a chemical formula 4 of R1X. R1 is (C1~C30)saturated alkyl, (C1~C30)alkenyl, (C1~C30)alkynyl, (C2~C29)alkylcarbonyl, (C6~C30)aryl of linear or basin-type. R2, R3, R4 and R5 are linear or basin-type of saturation or unsaturation(C1~C30) alkyl, (C6~C30)aryl, (C1~C30)alkyl (C6~C30)aryl, (C3~C30)cycloalkyl, alkoxy alkyl and (C1~C30)alkoxy(C6~C30)aryl. R6 is hydrogen, linear or basin-type of saturation or unsaturation (C1~C30)alkyl, (C6~C30)aryl, and (C1~C30)alkyl (C6~C30)aryl, (C3~C30)cycloalkyl, (C1~C30)alkoxy alkyl and (C1~C30)alkoxy(C6~C30)aryl. X is halogen atom, sulfate(SO4), sulfonic acid(SO3H), SO3R7, nitrate(NO3), phosphate(PO4) and ester(R7COO). R7 is (C1~C30)alkyl or (C6~C30)aryl.
