PROBLEM TO BE SOLVED: To provide a method for simply measuring the content of methylated DNA in the object DNA domain of a genome DNA contained in a biologic specimen, and the like. SOLUTION: This method for detecting or determining a methylated DNA has a first process of obtaining a single-strand DNA from a DNA sample originated from a genome DNA contained in a biological specimen; a second process for mixing (i) the single-strand DNA obtained in the first process with (ii) a methylated DNA antibody and (iii) an oligonucleotide which does not inhibit ligation of the methylated DNA antibody to a DNA methylated in the objective DNA domain and ligating to the single strand DNA to form a complex of the single-strand DNA, containing the object DNA domain with the methylated DNA antibody and the oligonucleotide, and simultaneously, or after forming the complex, the complex is separated, and the like.
