PROBLEM TO BE SOLVED: To provide a method for manufacturing, readily and industrially efficiently, a polysaccharide acylate by causing a polysaccharide or a not-wholly substituted polysaccharide derivative to react with an acid chloride, where the method gives a polysaccharide acylate excellent in hues and solvent solubility and having a low impurity content. SOLUTION: The manufacturing method of the polysaccharide acylate comprising causing a polysaccharide or a not-wholly substituted polysaccharide derivative to react with an acid chloride comprises (i) a step for adding an acid chloride to a solution of the polysaccharide or the not-wholly substituted polysaccharide derivative having a water content thereof adjusted to at most 1,200 wt. ppm to form the polysaccharide acylate and (ii) a step for adding an alcohol to the reaction mixture liquid obtained in the step (i) to esterify the remaining acid chloride.
