The present invention provides compositions for modulating translational repression and methods of use thereof. In particular, tethering constructs capable of mediating sequence-specific translation repression of target RNAs. The invention also provides method for enhancing miRNA-mediated translational repression in an extract, cell, cell lysate, or organism by introducing an effective amount of a sequestration polypeptide, an ortholog or bioactive fragment thereof, to the extract, cell, cell lysate, or organism. Preferred sequestration polypeptides are RCK, its ortholog or bioactive fragment. Therapeutic methods are also featured.
