摘要 A novel N-acetylglucosamine-2-epimerase is provided to mass-produce inexpensively CMP(cytidine monophosphate)-neuraminic acid by using inexpensive substrates including cytidine monophosphate and N-acetyl-D-glucosamine via one reaction. The CMP-neuraminic acid is produced by reacting a reaction mixture containing (i) cytidine 5' monophosphate(CMP), (ii) nucleotide triphosphate(NTP) or ATP, (iii) acetyl phosphate, (iv) N-acetyl-D-glucosamine and (v) pyruvic acid in the presence of (a) N-acetylglucosamine(GlcNAc)-2-epimerase having the amino acid sequence of SEQ ID NO:2, isolated from Bacteroides fragilis NCTC 9343, (b) N-acetylneuramic acid(NeuNAc) aldolase, (c) cytidine 5' monophosphate kinase, (d) acetate kinase and (e) CMP-N-acetylneuramic acid synthetase.
