<P>PROBLEM TO BE SOLVED: To provide improved vaccines. <P>SOLUTION: The improved vaccines are disclosed. The improved vaccines include a nucleotide sequence that encodes a coding sequence that comprises an immunogenic target protein linked to or comprising an intracellular cellular targeting sequence, and the coding sequence is operably linked to regulatory elements. Methods of immunizing individuals are also disclosed. The immunogenic target protein is a pathogen-derived antigen, a cancer-related antigen or an antigen related to autoimmune diseases.
