PROBLEM TO BE SOLVED: To provide a method for producing a tasty 5'-nucleotide useful for various seasonings including yeast extracts from a ribonucleic acid in a yeast extract in high yield. SOLUTION: A yeast extract containing ribonucleic acids is treated with a phosphodiesterase to decompose the ribonucleic acid to a 5'-nucleotide. In the above process, the yeast extract is preparatorily subjected to a depolyamination (especially a polyamine having≥3 amino groups in the molecule) treatment, a weakly acidic cation-exchange resin treatment or an adsorbing resin treatment.
