PROTOPLAST, ITS PRODUCTION AND METHOD FOR REGENERATING PLANT BODY FROM SAID PROTOPLAST

PURPOSE:To easily and stably obtain a protoplast capable of regenerating a plant body in high efficiency, by extracting a shoot anlage, removing the cell wall by enzymatic treatment and separating a protoplast. CONSTITUTION:A shoot apex of an arboreous or herbaceous plant (e.g. poplar or spinach) is extracted and subjected to spin culture to produce a shoot anlage. The shoot anlage is treated with an enzyme (e.g. pectolyase Y-23 or cellulase Onozuka RS) to remove the cell wall and then the protoplast is separated. The obtained protoplast is cultured in an artificial liquid medium composed mainly of phytohormone (e.g. naphthaleneacetic acid) and inorganic salts preferably in the presence of a supporting agent (e.g. gelrite) to form a callus, which is transplanted to a redifferentiation medium and the shoot is regenerated to effect the regeneration of the plant body.
