The invention relates to a method for detecting and/or identifying Escherichia coli (E. coli) in a biological sample, that comprises: a) inoculating the biological sample likely to contain E. coli on a detection medium that comprises tryptophane and a substrate of an A enzyme expressed by the majority of the E. coli in order to obtain bacteria colonies; b) detecting the colonies expressing the activity of the A enzyme and identifying them as E. coli; and c) detecting the colonies that do not express the activity of the A enzyme, carrying out an indole test, and identifying the colonies having a positive indole test as E. coli.
