E. COLI CAPABLE OF EXPRESSING CYTOCHROME P-450, AND BIOCONVERSION METHOD USING THE SAME
摘要
Disclosed is a means for improving the poor conversion efficiency in a bioconversion system using an E. coli cell having a bacterium-derived cytochrome P-450 gene integrated therein. A recombinant E. coli cell is produced by integrating genes aciB and aciB both encoding an electron transfer system derived from Acinetobacter sp. strain OC4 into an E coli cell, and adding a polynucleotide encoding an N-terminal sequence composed of 48 amino acid residues of AciA to the 5'-terminus of a bacterium-derived cytochrome P-450 gene, wherein AciA is an alkane-oxidized cytochrome P-450 derived from Acinetobacter sp. strain OC4. By using the recombinant E. coli cell in bioconversion, it becomes possible to convert a hydrophobic or amphipathic substrate compound into a desired compound with extremely high efficiency.
