PROBLEM TO BE SOLVED: To provide a method for isolating and purifying an amyloid plaque which is a specific pathologic structure of Alzheimer's disease from a brain tissue, to provide the plaque and a method for searching for a factor associated with the Alzheimer's disease and contained in the plaque. SOLUTION: The method for purifying the amyloid plaque comprises the following steps. (1) pulverizing the brain tissue, centrifuging the pulverized brain tissue at about 50,000 g in a buffer solution and separating pellets, (2) suspending the pellets in a buffer solution, adding the resultant suspension to an about 30% sucrose solution, centrifuging the resultant mixture at about 50,000 g and separating an amyloid plaque-containing fraction, (3) adding the separated amyloid plaque-containing fraction to an about 60% sucrose solution, centrifuging the obtained mixture at about 166,000 g and separating pellets, (4) suspending the separated pellets in a buffer solution and filtering the resultant suspension through a filter having about 40μm mesh, (5) separating pellets obtained by centrifuging the obtained filtrate at about 16,000 g and (6) adding the pellets to an about 10% sucrose solution, centrifuging the prepared mixture at about 4,000 g and separating the amyloid plaque-containing fraction.
