Provided is a transformed prokaryotic host cell comprising i) a chromosomal gene which inhibits cell growth, operably linked to a regulatory sequence encoding RNAII or a portion thereof; and ii) a plasmid comprising an origin of replication encoding an antisense sequence which is RNAI or portion thereof, wherein the binding of the antisense sequence encoded by RNAI to mRNA transcribed from RNAII inhibits the action of the chromosomal gene, thereby permitting cell growth. Further provided are similar prokaryotic host cells wherein the plasmid encodes an antisense sequence which is RNAI binding to mRNA transcribed from RNAI. Further provided are methods of maintaining the plasmid in a host cell, producing the plasmid, producing a recombinant protein using the host cells, pharmaceutical compositions comprising the host cells and use of the host cells in the manufacture of medicaments.
