A chitosan oligosaccharide is provided to induce quinone reductase or glutathione S-transferase activity excellently and inhibit the expression of cyclooxygenase 2, thereby being used as a cancer prophylactic agent of inhibiting rectal cancer development. A pharmaceutical composition for preventing rectal cancer comprises chitosan oligosaccharide as an effective ingredient. The composition further comprises a pharmaceutically acceptable carrier, an excipient and an additive.
