A composition comprising human immunodeficiency virus nucleocapsid(HIV NC) protein is provided to inhibit proliferation of human immunodeficiency virus through the overexpression of HIV NC, so that the composition is useful for preventing and treating AIDS(acquired immune deficiency syndrome). A composition for preventing and treating AIDS comprises a polypeptide comprising HIV NC protein having the amino acid sequence of SEQ ID NO:1, which is selected from SEQ ID NO:2 to SEQ ID NO:7. A HIV NC gene has the nucleotide sequence of SEQ ID NO:8. An expression vector contains a polynucleotide comprising the HIV NC gene, having the nucleotide sequence selected from SEQ ID NO:8 to SEQ ID NO:14. A method for screening a material useful for preventing and treating AIDS comprises the steps of: (a) co-culturing a recombinant cell expressing the polypeptide containing HIV NC protein and a candidate material; and (b) measuring the increase of polypeptide level in the cell.
