<P>PROBLEM TO BE SOLVED: To provide a technique etc., of detecting the initiation of primer extension with fluorescence in real time system. <P>SOLUTION: There is provided an oligonucleotide having a quencher Q bound to the 3'-position of a pentose of a 3'-terminal nucleotide residue N<SB>1</SB>and a fluorescent substance F bound to a nucleotide residue N<SB>2</SB>present within a range where the quenching function of the quencher Q attains. In the oligonucleotide, a base of the 3'-terminal nucleotide residue N<SB>1</SB>is a mismatched base for a template DNA.
