A method of detecting pathogenic aggregates of a protein in a sample comprises contacting the sample with a first monoclonal antibody or epitope binding fragment thereof, the first monoclonal antibody or an epitope binding fragment thereof being immunoreactive with the protein of the pathogenic aggregates, removing pathogenic aggregates of the protein not bound to the first monoclonal antibody or epitope binding fragment thereof, contacting the bound pathogenic aggregates of the protein with a second monoclonal antibody or epitope binding fragment thereof, the second monoclonal antibody or epitope binding fragment thereof being substantially identical to the first monoclonal antibody or epitope binding fragment thereof and being coupled to a detectable amplification module, amplifying the amplification module coupled to the second monoclonal antibody bound to the pathogenic aggregates, and detecting the amount of amplified amplification module to determine the presence of protein aggregates in the sample.
