The aim of this invention is to offer a kind of chromatographic cake and its manufacturing method applicable to the separation and simultaneous renaturation and purification of biopolymers with excellent effect. Said chromatographic cake includes a chromatographic packing cake with the mobile phase inlet and outlet and a chromatographic packing packed in the inner cavity of the chromatographic packing cake. The ratio of thickness and the radial length of the inner cavity of the chromatographic packing cake are smaller than or equal to 1.
