PROBLEM TO BE SOLVED: To provide a method for preparation of a marker for discriminating mushroom belonging to genus Pleurotus. SOLUTION: The marker is prepared in the following processes, allocating genes by a restriction enzyme from all DNAs of the sample Pleurotus eryngii, isolating fragments including CT microsatellite region from the all fragments, attaching adaptors on its outside and inserting it into a plasmid vector, amplifying the plasmid by transformation into E.coli, specifying a colony having plasmid including the CT microsatellite region by a colony hybridization, picking up the DNA fragments from the colony, and amplifying DNA fragments including the CT microsatellite region in the DNA fragments by using a T7 primer and SP6 primer, obtaining a DNA base sequence on both sides of the CT microsatellite region and then designing a primer specifically amplifying the CT microsatellite region from the base sequence.
