A uniflow scavenging micro-engine and a method of operating the micro-engine are provided to increase efficiency by realizing uniflow scavenging and thereby reducing the loss of a mixture of fuel and air, and to increase thermal efficiency through combustion by initial ignition using a glow plug and subsequent compression ignition. A uniflow scavenging micro-engine is operated by a fuel transmitted from a DME fuel tank. The micro-engine includes inlet parts(1,8), glow plugs(3,6), and a combustion chamber(4). The inlet parts are provided on opposite ends of the micro-engine to introduce a mixture of the fuel fed from the DME fuel tank and air. Each glow plug is layered on one side of the corresponding inlet part and ignites the mixture using a heater coil. The mixture, ignited by the glow plugs, explodes in the combustion chamber, thus sliding a piston(5) provided in combustion chamber leftwards and rightwards.
