A protein chip is provided to detect a disease marker with high sensitivity by comprising a self-assembled protein chip which is an active type probe having constant direction property on a template surface and improve integration degree of the probe per unit area, thereby being usefully used for developing a diagnostic kit. A protein chip is characterized in that a protein nano-particle where a disease marker specific epitope is linked with a ferritin heavy chain is integrated into a template surface. A method for detecting a disease marker based on PCR using the protein nanoparticle comprises the steps of: (a) obtaining a sample from a test body; (b) treating the protein nanoparticle integrated template surface with the sample and washing it; (c) treating the template surface with a secondary protein nanoparticle where a polylysine tag is linked with the protein nanoparticle and washing it; (d) treating the template surface with a first DNA primer and washing it; and (e) after adding a PCR reaction solution including a second DNA primer, a thermostable DNA polymerase, and a template DNA to the template surface, subjecting it to a PCR reaction. A diagnostic kit comprises a secondary antibody quantum dot coupled to the protein nanoparticle where the disease marker specific epitope is linked with the ferritin heavy chain or a DNA absorbed secondary nanoparticle and the protein chip.
