A mammalian cell comprising: a first reporter gene construct (RGC) comprising a nucleic acid (NA) molecule encoding a fluorescent protein-centromere protein (preferably CENP-A) fusion, operably linked to/under control of a constitutive promoter; a second RGC comprising a NA comprising one or more expression controls sequences (ECS), preferably DNA damage induced promoter GADD45a, operably linked to a nitroreductase (NTR) enzyme sequence. Also claimed is: a process for creating a cell line; a diagnostic kit; a method for determining the effect of a test agent on the phenotypic property of a cell comprising: providing cells comprising: said first RGC wherein said fluorescent protein is capable of emitting light of wavelength g 1; said second RGC; a DNA staining dye that emits light of wavelength g 2 when bound to DNA; and contacting said cells with: a test agent or control; NTR substrate (preferably a squaraine or cyanine dye) capable of emitting light at wavelength g 3; measuring intensity of g 1, 2 and 3 wherein a difference between test and control is indicative of a phenotypic effect. |
