A primer for detecting Potato spindle tuber viroid(PSTVd) is provided to diagnose the infection of the PSTVd from a small amount of a sample of in vitro culture plantlets or nutriculture and detect the PSTVd accurately due to no specific reaction with other main viruses. A primer for detecting Potato spindle tuber viroid(PSTVd) consists of a sense PSTVdf(5'-GAACTAAACTCGTGGTTCCTGT-3') described as SEQ ID : NO. 1 and an anti-sense PSTVdr(5'-GTTCCAAGGGCTAAACACCCT-3') described as SEQ ID : NO.2.
