A composition containing a swellfish extract using protease and its preparation method are provided to increase the amount of effective ingredients contained in the swellfish extract and obtain the effect of enhancing liver function by adding Cynara scolymus powder. A composition containing a swellfish extract using protease is prepared by the steps of: (i) separating flesh from swell fish; (ii) allowing 0.1~0.2% of protease to react with the flesh of swellfish at 25~50°C for 12~36hr; and (iii) distilling the reaction product at 100°C or below to obtain a distillate. The step (iii) further comprises adding Cynara scolymus powder to the distillate at a level of 2~8g/L, filtering it with a filter of 1000~1400mesh, and aging the filtrate at 40~60°C for 5~20hr.
