A phone number-based e-mail interworking messaging service method in a mobile communication system, and a system therefor are provided to allow a user to conveniently send an e-mail through a phone number of a recipient, and increase a usage rate of sending e-mails by using a mobile terminal. A database server(150) stores phone numbers of recipients by matching them to e-mail addresses. If a phone number of a recipient of an MMS(Multimedia Messaging Service) message received from a sender does not have an e-mail identifier, an MMSC(MMS Center)(130) transmits the MMS message to the phone number of the recipient, and if the phone number of the recipient has an e-mail identifier, the MMSC searches an e-mail address mapped to the phone number of the recipient from the database server, and transmits the received MMS message to an external e-mail server(190) corresponding to the searched e-mail address.
