AN HBV VACCINE AND A PROCESS OF PREPARING THE SAME

A hepatitis B virus(HBV) vaccine is provided to induce excellent immunogenicity and strong cellular immunity by providing various epitopes, thereby being usefully used as a therapeutic vaccine of the HBV. An HBV vaccine comprises: a recombinant large surface antigen of HBV(LHBsAg) which consists of three kinds surface proteins of an L protein, an M protein and an S protein of the HBV and is located on the surface of a particle formed by binding between a preS antigen consisting of a preS1 and a preS2 and an S antigen; and a core antigen of the HBV. An HBV vaccine composition comprises the HBV vaccine and an immunological adjuvant. A method for preparing the HBV vaccine comprises the steps of: (a) introducing a 3'-UTR nucleotide including an encoding region of an envelope gene of the HBV and a polyadenylation portion into a pMSG vector(deposition no. KCCM 10202); (b) tranducing the expression vector prepared from the step(a) into an animal cell; and (c) culturing the transduced animal cell to obtain a recombinant large surface antigen(L-HBSAg) protein.
