PROBLEM TO BE SOLVED: To provide a method for producing a variant with deleted or substituted wide gene region on a chromosome of a mold constituting a gene cluster relating to biosynthesis of aflatoxin, and to provide a variant having deleted wide gene region. SOLUTION: The variant is prepared by making a cassette by connecting two sequences apart more than some kilobase on a chromosome of an organism by mediating a selective marker gene sequence in the center, and then transforming it into cells to delete or substitute the wide gene region. The selection of the variant is carried out in the following processes, selecting organism with a gene transduced on the chromosome by utilizing the characteristics of the selective marker, a process directly inspecting changes of gene structure on each variant to select the objective variant with deleted gene region and substituted gene region.
