An agent for preventing and treating prion diseases is provided to treat the prion diseases and all diseases related to damage thereof and prevent the prion diseases conveniently. An agent for preventing and treating prion diseases comprises at least one selected from the group consisting of a cationic amino acid monomer, a cationic amino acid polymer and a stereoisomer, at least one selected from the group consisting of a polyanionic compound and a homologue thereof, and MgCl2, wherein the amino acid is selected from the group consisting of lysine, arginine, histidine and ornithine, the polyanionic compound is selected from the group consisting of phosphotungstic acid, sodium tungstate, sodium molybdate, ammonium paramolybdate, and uranyl acetate, the selected cationic amino acid monomer is 5-40 mM, the concentration of the cationic amino acid polymer and the stereoisomer is 1-10mug/ml, the concentration of the polyanionic compound is 0.001-0.008% based on 8.5mum of the MgCl2, and the amino acid polymer used is about 2mug/ml of an L-lysine polymer having a molecular weight of more than 30KDa, or about 1-1.5mug/ml of a D-lysine polymer having a molecular weight of 15-17KDa, or about 2.5-5mug/ml of an L-lysine polymer having a molecular weight of more than 15KDa.
