Polypeptide, having (R)-hydroxynitrile lyase activity (I) is new. Polypeptide, having (R)-hydroxynitrile lyase activity, (I) that is: (a) a 536 amino acid (aa) sequence (S1) defined in the specification; (b) a non-glycosylated 532 aa sequence (S2) defined in the specification; (c) a sequence at least 60% identical with (S2); or (d) a non-glycosylated sequence that represents a fragment of (S2). Independent claims are also included for the following: (1) polynucleotide (II) that: (a) is a 1593 bp sequence (S3) defined in the specification; (b) encodes (S1); (c) is at least 95% identical to sequence (S3); (d) is mutated but at least 60% identical to (S3); or (e) the complement of (a)-(d); (2) plasmid or vector that contains (II); (3) cell that contains the plasmid or vector of (2) and/or (II); (4) method for producing (I) by culturing cells of (3); and (5) method for preparing cyanohydrins (III) by reacting a carbonyl, or related, compound with a cyanide donor in presence of (I).
