发明名称 METHOD FOR MEASURING HELICASE ACTIVITY
摘要 PROBLEM TO BE SOLVED: To provide a method for capable of measuring the activity for dissociating the double-stranded nucleic acid of helicase to a single-stranded nucleic acid, simply and at low cost by using neither a radioactive isotope element, an electrochemical measuring means nor a FRET. SOLUTION: This method for measuring the helicase activity is provided by preparing a fluorescence-labeled double-stranded nucleic acid as a substrate for measuring the helicase activity, by using a nucleic acid labeled with a fluorescent pigment changing its fluorescence intensity, by the approaching and leaving of nucleic acid-constituting bases and another nucleic acid having a base sequence complementary with the nucleic acid; conducting enzyme reaction by the helicase, by using the fluorescent-labeled double-stranded nucleic acid as the substrate; and measuring the changing fluorescent intensity that accompany the dissociation into the single-stranded nucleic acid.
