PROBLEM TO BE SOLVED: To provide an absorption medium having capacity of same or improved IgG bond, compared with a well-known variant having (a) IgG bond specificality of a protein A, (b) at least same degree of stability as that of other adsorbent using a natural protein A as a base and (c) a matrix-binding rProtein A-cys. SOLUTION: The present invention provide a separation medium comprising a base matrix substituted with a group complying with formula I [wherein (a) rProtein A-cys is a recombinantly produced Protein A which contains cysteine in its amino acid sequence; (b) B is a bridge which binds to the base matrix and (c) X contains a heteroatom N or S originating from rProtein A-cys].
