A transposable construct comprises at least four inverted repeats, at least two of which are inverted in relation to the others, comprises a DNA of interest located between two pairs of opposing repeats excisable be a transposase in situ, to allow removal of the flanking transposon-derived repeats from either side of the DNA of interest following insertion into the genome, thereby preventing further transposition. The construct is preferably derived from the insect PiggyBac transposon, and is preferably used to transform an insect. A further transposable element comprises at least three inverted repeats, at least one of which is inverted in relation to the others, wherein at least one non-terminal repeat is a minimal repeat. A DNA of interest may be located between the minimal repeat and one of the terminal repeats, and recombination between the minimal repeat and the other terminal repeat prevents further transposition. |
