GELDANAMYCIN DERIVATIVES BY MODIFICATION OF BIOSYNTHETIC GENES

A geldanamycin derivative is provided to show Hsp90 inhibitory action similar to geldanamycin, thereby being usefully used as an antibiotics, an anti-fungal agent, an anti-viral agent, an immunosuppressive drug, a degenerative neuro-disease therapeutic agent, an anti-inflammatory agent or an anti-cancer agent. A geldanamycin derivative is represented by the formula(1), wherein R1 is OH or OCONH2, or by the formula(2), wherein R1 is H or OH, and R2 is OH or OCONH2. A recombinant vector includes a gene construct double inactivated by gel7 and gel16 or gel7 and gel8. An antibiotics, anti-fungal agent or anti-viral agent comprises the geldanamycin derivative, the recombinant vector, a mutant strain of Streptomyces hygroscopicus subsp. duamyceticus, and a geldanamycin derivative produced by the mutant strain.
