<p>The invention relates to a process for the enzymatic preparation of 6-O-acylsucrose (Figure 1) or a derivative thereof
in the presence of at least one non-nucleophilic organic solvent, by selective monoacylation of sucrose on the 6-position with a donor acyl ester of formula R 1 R 2 C=CR 3 -O-C(O)R, in which R 1 , R 2 , and R 3 each independently represent hydrogen, an optionally substituted (hetero)alkyl group or an optionally substituted (hatero)aryl group with for example 1-50 C atoms, or of formula R 4 -O-C(O)R, in which R 4 is a halogenated alkyl group of 2-50 carbon atoms in which at least the 2-position of the alkyl group is substituted with one, two or three halogens, and with R representing an optionally substituted (hetero)alkyl or (hetero)aryl group having from 1 to 50 carbon atoms, wherein the acylation is carried out in the presence of a lipase enzyme selected from the group of
(i) lipases obtainable from a micro-organism from a Surkholderia gladioli species; or
(ii) any such Burkholderia gladioli lipase enzyme produced and over-expressed in another suitable micro-organism; or
(iii) mutants of the lipases from (i) and/or (ii) having selective acylation activity to form 6-O-acylsucrose in acylation of sucrose.</p> |
