METHOD FOR SCREENING ESSENTIAL METABOLITES IN GROWTH OF MICROORGANISMS

A method for screening essential metabolites in growth of microorganisms is provided to improve screening convenience of the essential metabolites and genes associated with them, screen materials inhibiting the growth of microorganisms, and anticipate drug target genes of pathogenic microorganisms by deleting the genes associated with the essential metabolites. A method for screening essential metabolites in growth of microorganisms comprises the steps of: (a) selecting a target microorganism and constructing the metabolism cycle model of the selected microorganism; (b) fixing the metabolite flow value of the consumption reaction equation for each metabolite at zero(0), calculating the reduction of cell growth rate caused by a metabolite by using a mathematical equation 2: F = (r_loss)/(r_wild) in which r_wild is an original cell growth rate, and r_loss is a reducing cell growth rate when the corresponding consumption reaction equation is inactivated, and deciding the metabolite as an essential metabolite in growth of microorganisms when the cell growth rate treated by the metabolite is reduced by 50% or less as compared with the original cell growth rate; (c) defining the availability of the metabolite selected in the step (b) as flux sum in a mathematical equation 3, and calculating the flux sum of each metabolite; and (d) confirming the metabolite as an essential metabolite in growth of microorganisms when the metabolite induces reduction of flux sum, i.e. induces reduction or termination of the cell growth. Further, the target microorganism is a colon bacillus or pathogenic microorganism.
