An apparatus and a method for canceling interference in a broadband wireless access communication system are provided to be applicable to a high speed fading environment and enhance the performance of canceling an interference component. A preamble channel estimating unit(430) calculates channel information of a preamble of a received signal. An initial weight value calculating unit(440) calculates a weight value of a preamble interval based on the preamble channel information. With reference to the first symbol of traffic, a weight value updating unit(450) updates a weight value for canceling an interference of a traffic symbol based on the weight value calculated by the initial weight value calculating unit(440), and with respect to the other remaining symbols of the traffic, the weight value updating unit(450) calculates a weight value of the symbols of the traffic and updates a weight value for canceling an interference of the traffic symbols. An integrating unit(460) receives the initial weight value and the updated weight value, and cancels an interference from a reception signal. A decoding unit demodulates and decodes the interference-canceled signal according to a previously defined modulation/demodulation level.
