The gels are prepd. by (1) suspending a soln. of PVA (in water, opt. mixed with organic solvent for PVA) in a PVA - immiscible organic phase without addn. of emulsifier, and (2) during the suspension process, crosslinking by acid-catalysed reaction with an added dialdehyde. Pref. the PVA soln. has viscosity 5-200 cP and the organic suspension phase has viscosity 10-200 cP (both measured at 20 deg. C). Pref. the organic phase is a plant, paraffin or mineral oil and/or (un)satd. fatty acid. It may have 1-20 vol.% of an organic solvent immiscible with the polymer phase. PVA pref. has deg. of polymerisation 300-6000. USE/ADVANTAGE - Used for affinity chromatography support and for haemodiffusion, esp. to separate from human blood (prod. blood gp. antibodies; lysosomal enzymes (for treating kidney disease) or low density lipoprotein (LDL; for treating arteriosclerosis or hypercholesterolaemia). The beads may also be used in immunoassays. By controlling the viscosity of the organic phase (to 30-80 cP) the use of emulsifier, electrolyte additives and volatile organic solvents may be avoided. The prod. porosity may be controlled by adjusting the dialdehyde concn. and/or the deg. of polymerisation of PVA, opt. also by addn. of organic solvent. Particles of size 1-3000 microns may be made from the same starting material, and the size is controlled by stirring speed and viscosit |
