A method for microclonal propagation of hop regenerants grown from apices in vitro consists in that in sterile conditions one hop regenerant is divided into micrografts according to the number of internodes, the length of lamina is shortened, a nutrient medium is formed with micronutrients according to the formulation of Murasige and Skoog, macronutrients, vitamins as thiamine-HC, pyridoxine-НСand mesoinosite, a source of carbohydrates, phytohormones as- indoleacetic acid, agar-agar. The micrografts are made with sizes of 10- with a lamina length of 0.5-1.0 of the length of a micrograft. A nutrient medium is formed with macronutrients according to the formulation of Niche and Nich. The vitamins are supplemented with folic acid in an amount of 1-5 mg/l, and the thiamin-НСis used in an amount of 1-5 mg/l, pyridoxine-НСof 1-5 mg/l, mesoinosite of 80-120 mg/l. Glucose in an amount of 20-40 g/l is used as the source of carbohydrates. Kinetin in an amount of 0.05-0.5 mg/l and-indoleacetic acid in amount of 6-15 mg/l are additionally used as the phytohormones. The micrografts are grown on the nutrient medium at a temperature of +23-27 °С, an illumination intensity of 1.5-3 kLx, at a 14-17-hour light period up to forming a rooted microseedling. Then the process is repeated up to producing the necessary number of the microseedlings, which are planted into a peat and sand mixture in a ratio of 3:1-3:2 with pH 5.5-6.5, a moisture content of 40-70 %, preliminary removing the remains of the nutrient medium from the root system and carrying out the treatment of the latter in a fungicide. |
