A composition comprising high-molecular weight of hyaluronan is provided to inhibit differentiation of osteoclasts by inhibiting M-CSF(macrophage colony-stimulating factor) signaling and RANKL(receptor activator of NF-kappaB ligand) signaling in osteoclast precursors, so that the composition is useful for inhibiting bone resorption. A composition for inhibiting bone resorption comprises hyaluronan having molecular weight of 1,000-10,000 kDa which inhibits osteoclastogenesis by binding with TLR4(toll-like receptor 4), reduces RANK expression by inhibiting activation of AP-1(activator protein 1) and MITF(microphthalmia-associated transcription factor) through inhibition of M-CSF signaling osteoclast precursors, and blocks Rac activity, ROS(reactive oxygen species) generation and MAPKs(mitogen-activated protein kinases) activation by M-CSF in osteoclast precursors, wherein the composition prevents and treats disease associated with bone resorption including osteoarthritis, osteoporosis, metastatic carcinoma or Paget's disease.
