PROBLEM TO BE SOLVED: To provide a fluorescence image acquisition device and a fluorescence image acquisition method capable of acquiring suitably a fluorescence observation image of a sample with high resolution. SOLUTION: This fluorescence image acquisition device has a configuration equipped with a sample stage 15, an image acquisition part 30 including an imaging device 31 for acquiring an image using a prescribed direction as a longitudinal direction, a fluorescence optical system unit 44 including a dichroic mirror 45 for vertical illumination by excitation light from an excitation light source 40, a scanning part for acquiring a partial image by scanning the sample S by the imaging device 31 repeatedly in a plurality of times, an image synthesis part for generating the fluorescence observation image of the sample S by synthesizing a plurality of partial images, a correction data storage part 76 for storing shading correction data in correspondence with the optical system unit 44, and an image correction processing part 75 for performing shading correction to each of the plurality of partial images on reference to the shading correction data.
