A plasmid pILR091 isolated from lactobacillus capable of being easily settled in the intestine of porcine is provided to have both advantages of an RCR plasmid and a theta-replication plasmid and show no big change of the replication numbers for succeeding 20 generations. A method for preparing a transformant using the same is provided to obtain the transformant used for preparing an oral carrier of the porcine, thereby significantly increasing the potential energy and possibility of probiotics. A plasmid pILR091 includes a sequence described as SEQ ID : NO. 1 and is isolated from Lactobacillus reuteri L09(deposition no. KACC 91266P). A shuttle vector includes the plasmid pILR091; a replication origin of E. coli; a promoter; an MCS(multiple cloning site); and a selection marker, where the promoter is selected from the group consisting of CMV(cytomegalovirus) promoter, CaMV(Cauliflower Mosaic Virus) 35S promoter, rplL(recombinant porcine interleukin) promoter and ptsH(phosphotransferase system HPr(phosphocarrier protein)) promoter. A method for transforming comprises a step of introducing a recombinant vector where a target gene is inserted into the MCS of the vector into bacteria.
