A method for mass-producing FK506(tacrolimus) using a fkbR1 introduced strain is provided to increase the production amount of the FK506 about 3 times higher than a mother strain, thereby being very useful for biological medicine industry. A method for mass-producing FK506(tacrolimus) comprises the steps of: (a) transforming a host cell using a recombinant DNA where a fkbR1 as a gene encoding a biosynthesis controlling enzyme of FK520(ascomycin) derived from Streptomyces hygroscopicus var. ascomyceticus ATCC 14891 strain is introduced; and (b) culturing the host cell.
