A fuel cell and a manufacturing method thereof are provided to stably seal an anode separator and a cathode separator, and to save a production cost by reducing the amount of an adhesive used. A fuel cell(200) includes: many unit cells(220) comprising a membrane-electrode assembly(221), a cathode separator(240) which is adjacent to one side of the membrane-electrode assembly and has oxidant channels(247), and an anode separator(230) which is adjacent to the other side of the membrane-electrode assembly and has fuel channels; and fastening plates(253) disposed on the outmost sides of the unit cells. At least one separator of the anode separator and cathode separator has coolant channels(231) and a groove(232) disposed to adjoin the coolant channels, wherein the groove is injected with an adhesive. The other separator to come in contact with the one separator having the groove has a protrusion which is formed in a part corresponding with the groove and is inserted into the groove.
